I recently got a DSLR, Nikon D80. After reading different threads I am still struggle to understand exposure related techniques. Many members suggest that to get the proper exposure, use spot meter and in M mode, lock the exposure. I am really not getting that. My user manual says AE doesn't have any effect on M mode, page 53. Is there any technique to lock the exposure in M mode? Please advice.
Also I check the histogram after the shot. Is there any mechanism to analyze the histogram before the shot?

First, by shooting in M mode, you are locking the exposure. You're setting both aperture and shutter speed, which effectively locks the exposure (if you're not changing ISO).
The design of (nearly all) DSLRs requires that an image be taken in order for histogram data to be available. When the viewfinder is in use, the mirror id in the "down" position, which blocks light from the sensor. Thus, no LCD preview, and no pre-shot histogram is possible.
