I'm wondering if I'm only allowed to take a single exposure(JPG/RAW), should I expose it for hightlights or shadows and later corrected the under/overexposed areas in Photoshop? I'm particularly interested in the reasons. Thanks!

Expose to place the brightest part of the scene at the tone you want it in the final image, or at least as close as you can get it. RAW adjustemnts to tone are better, but works well with jpeg also.

$64,000 question.
I prefer to set metering on spot or central 9% as it is on my 350D, press the shutter 1/2 halfway to turn on metering and scan the scene I am shooting. I generally find my brightest area, lock the exposure, re-compose and shoot.
I can bring back the shadows, but I can't fix blown-out highlights.

I do the same, but I lock it with a +1 to +2 EV compensation to keep it from underexposing everything.
