This isn't just for kirbic, but I think he knows the answer:

If I wanted to use the canon 2x extender + extension tube + 50mm 1.8

How long of an extension tube would I need? Would that length extension tube work no matter what lens I used (that is, non 2x extender ready lens)?

Also, this doesn't change the focusing distance, does it?

You can use a 12mm extension tube, which is the shortest available. Like any extension tube, it will also enable the lens to focus closer, but you will also lose infinity focus. Using a longer tube (or stacked tubes) will further reduce the minimum focus distance and increase magnificitation ( I have not tried anything but the 12mm tube).
I just tried mounting the 2.0x II, EF-12 extension tube, and 50/1.4 lens. I found that the range of focus is quite limited, about 8 inches when the lens is focused to infinity, and about 4 inches when the lens is set to closest focus.
