Once I know where you'll stay I'll tell you precisely how to get there form the airport.
I recommend this inexpensive one (the cab will be around $26 +tip):

M60 bus to 125th Street in Manhattan (follow signs at the airport or ask at info where is the bus). $2.25 bus fare (available 4:50 a.m. - 1 a.m.). Transfer is available to 1/2/3, 4/5/6, A/C and D subway trains (free transfer with MetroCard, otherwise $2 additional).

First thing is to get a Metrocard let's say for $20 and is good for bus/subway with transfer within 2 hours.

I am surprised you did not a better deal with expedia, air and hotel.

He's calling me back today to complete the booking. The address of the hotel is 21 West 37th Street. Google maps shows it as being really close to Time Square. Booked my flight through Delta. Cheapest I could find. Believe it or not but this just seemed to be the right path for me. You think you get deals with these travel sites but it's not always the case. When you go to finalize what you thought was a good deal they add tax and fees onto that and it puts it about the same price that I'm paying. Plus this hotel seems to be really close to everything which should make life easy and fun.

Is this Metrocard for $20 good for bus and Subway the entire time we are there? One day? I'm going to contact the hotel also to ask if they provide free shuttle?

When you say 1/2/3....are those streets you are referring to?

Also, if you could tell me the best ways to travel to:
Staten Island Ferry
Ellis Island
Central Park. (Seems that you can walk to it?) BTW I'm used to waling many blocks without issue.
World Trade Center
Rockefeller center
Brooklyn bridge

I think these are the main sites on our list to try to get in.

I answer you quickly but will make a little package for you, info, maps, coupons, etc. Is this in August?
- one bus/subway tkt is $2.25 transfer between them within 1 hour, if it's between bus and bus should be in perpendicular direction or different directions not same bus downtown and same uptown. Easy.
- if you buy a $10 or $20 metro card they give you extra cents or $1.21 (peanuts) as a bonus. Can use same card for more people and it takes the fee or gives the transfer for each one. There are deals for 1 day pass good if you plan to go more than 5 times one day. Buy at any subway entrance.
- walking is the best thing. Mighty crowded where you stay, but that's Midtown's definition
- rule of thumb: 10 blocks = 10 minutes (a lot more for window shopping and photo). 20 blocks = 1 mile. We walk A LOT here. For me personally, the average day I walk at least 4 miles just to do my things.
- from where you are , 30' to Central Park not rushing
- you'll be by the Empire State Bldg (34th street)
- walk Uptown on 6th Ave to Bryant Park and Fifth Ave, Public Library (go inside)
- could certainly walk all the way Downtown, very pleasant on Bway or Fifth Ave and stumble upon Flatiron Bldg (Stieglitz). You can walk uptown on Bway also and see Lincoln Center @ 64th str.
The beauty is to discover and walk the streets. Walk up on Madison from 50th str for fashion.
A LOT to see Downtown.
Go on Google and put the address and look around (the views are a bit old with a lot of contructions) but you get a feeling.
