Will it work to stick a Canon 500D close up filter on a Canon EF 100mm f/2.8 USM Macro lens?

In theory it should have the same effect it has on other lenses, to shorten the minimum (and maximum) focusing distances. That would make the minimum distance *really* short, and shots pretty hard to light.
If you have one, it doesn't hurt to try it. If you don't have one, it's an expensive test. If you don't have one, post back and let me know, I can test the combination, though I'd have to manually hold the 500D up to the lens (my 500d is 70mm, and I don't have a 55-78mm step ring).

Interesting. Never thought to try because I didn't think you could. I have a 500D and a 50mm close-up lens on my Olympus. If I remember I will try it out in the next few days and let you know also.

Reason for asking is that I have 100mm Macro lens and there is a 500D up for sale, and I wonder if this combination would give me more than 1:1 macro or jsut very short focusing distance.

So I would be very thankful if you could run a quick test and tell me about the outcome of it.

Well, took me a while longer than I planned to get to this, but I was able to test the 500D with the 100/2.8 Macro. It works, and the working distance isn't *too* obscenely short. It seems to increase magnification by a factor of about 1.2 or 1.3, so not a huge gain in magnification.

Thank you for your effort, you saved me from purchasing a gadget that I don't have a need for at the moment.
